南极企鹅外周血细胞初步研究

摘要:
2012年8月至10月,在天津海昌极地海洋世界采用Wright-Giemsa’s染色和Natt-Herrick's染色方法研究了南极企鹅(
Pygoscelis
antarcticus)外周血细胞。研究表明，外周血液分类为：红细胞、白细胞和血栓细胞。其中白细胞包括：异嗜性粒细胞、嗜酸性粒细胞、嗜碱性粒细胞、淋巴细胞和单核细胞。测得红细胞平均大小（长径短径）为15.32μm×7.83μm，异嗜性粒细胞直径为(11.98±0.97)
μm，嗜酸性粒细胞直径为(11.61±0.78) μm，嗜碱性粒细胞直径为(11.87±0.69)
μm，淋巴细胞直径为(9.93±1.29) μm，单核细胞直径为(15.04±0.93)
μm，血栓细胞直径为（6.96±0.79）μm。统计红细胞总数为(1.85±0.17)×1012/L，白细胞总数为(6.37±1.68)×109/L。

A Primary Study on
Peripheral Blood Cells of Pygoscelis
antarctica

Abstract: From August to
October in 2012, this research provides a guide for study on
hematology of Pygoscelis antarctica in Tianjin Haichang
Polar Ocean World. The samples of peripheral blood was collected
from Pygoscelis Antarctica and was examined by
Wright-Giemsa’s stained smears with light microscope. Three
types of blood cells were encountered as follow: erythrocyte,
leukocyte and thrombocyte. Leukocytes that were recognised included
heterophils, eosinophils, basophils, lymphocytes and monocytes. The
erythrocytes mean size was 15.32μm×7.83μm, the mean diameter of
heterophils was 11.98±0.97 μm, the mean diameter of eosinophils was
11.61±0.78 μm, the mean diameter of basophils was 11.87±0.69 μm,
the mean diameter of lymphocytes was 9.93±1.29 μm, the mean
diameter of monocytes was 15.04±0.93 μm, the mean diameter of
thrombocytes was6.96±0.79μm. Dilute the peripheral blood with the
Natt & Herrick's solution. After discharged into the
hemacytometer counting chamber, count the total number of red and
white blood cells by light microscope. The erythrocyte counts were
(1.85±0.17)×1012/L, and the leukocyte counts were
(6.37±1.68)×109/L.

企鹅是鸟纲、楔翼总目(Impennes)所有种类的通称。主要生活在地球的南半球，目前已知全世界的企鹅共有18种。近年来，随着我国旅游业的不断发展，全国各地兴建近80家水族馆、海洋世界。珍稀的企鹅是极地环境下的代表动物，其憨态可掬的外形受到广大游客的欢迎，因此各个海洋馆拟引进极地企鹅作为建设场馆的重点项目之一。我馆引进的企鹅品种为南极企鹅（Pygoscelis
antarctica）又称帽带企鹅、颊带企鹅、胡须企鹅，属阿德利企鹅属(Pygoscelis)，因有一条黑色细带围绕在下颚而得名，主要分布在南极半岛北端西岸的南雪特兰群岛及亚南极岛屿。
Bonatt等[1]研究了阿德利企鹅属企鹅的外周血红细胞形态，并测得红细胞直径为(15.47±0.20)μm。目前，国内尚未有企鹅血液相关研究报道。为此，笔者对南极企鹅的血细胞进行显微镜观察，对其血细胞形态、数量、大小、白细胞分类等参数进行了观测和研究,旨在为相关人员在企鹅血液常规检测方面提供借鉴，进而有利于企鹅的健康状况评价和疾病诊断。

材料与方法
1.1 材料
1.1.1 动物来源
南极企鹅12只，6♀6♂，体重3.5kg~4kg。饲养环境为温度﹣5℃~﹣2℃，湿度60%~70%，水温3℃~4℃。试验动物均来自天津海昌极地海洋世界。
1.1.2 样品采集
常规饲养条件下观察无异常，依据Sharon的方法[2]，每只企鹅采血前空腹12h，由饲养员保定，碘伏-酒精消毒翅膀内静脉表面皮肤，后翅静脉采血3ml，装入含EDTA-K2抗凝剂的采血管内，充分摇匀，以待检测。
1.2 方法
1.2.1 外周血细胞计数
采集新鲜的静脉血后，通过Natt-Herrick's染液[3]对血液稀释和染色，用血细胞计数板在显微镜（OLYMPUS
CX31）40倍镜观察下统计红细胞和白细胞的总数。
1.2.2 外周血细胞显微结构观察
以新鲜静脉血液制作血涂片，用Wright-Giemsa’s染液染色，在显微镜（OLYMPUS
CX31）油镜（100倍）下进行血细胞的形态学观察和拍照。血细胞大小的测量以每只企鹅的血涂片在油镜的观察下，用测微尺随机测量100个血细胞的长径和短径以及细胞核的长径和短径的均值为测量结果。
1.2.3 血红蛋白含量与红细胞压积测定
将采血管内的血液充分摇匀后，进行血红蛋白含量与红细胞压积的检测，采用血常规检测仪（NIHON
KOHDEN MEK-6410C）读取结果。
2 结果
血液指标测定结果见表1。

2.1 红细胞
南极企鹅的成熟红细胞与鸟类的红细胞基本一样，形态为椭圆形、双面凸出，并且最主要的特征为内含细胞核。红细胞平均大小（长径×短径）为15.32μm×7.83μm。细胞核呈椭圆形，位于细胞中央，平均大小（长径×短径）为5.90μm×2.96μm。经染色后细胞质呈淡红色，细胞核被染成深蓝紫色，细胞核中染色质密集(图版I-A)。在血涂片中偶尔可见晚幼红细胞，略呈圆形，细胞质嗜碱性，故染色偏蓝(图版I-B)。

2.2 白细胞
跟其他鸟类一样，南极企鹅的白细胞可分为异嗜性粒细胞、嗜酸性粒细胞、嗜碱性粒细胞、淋巴细胞和单核细胞5种。白细胞分类统计百分比见表3，异嗜性粒细胞在白细胞分类中占比例最大。

2.2.1 异嗜性粒细胞
异嗜性粒细胞在白细胞中占(75.84±8.26)%，细胞直径为(11.98±0.97)
μm。细胞近似圆型，视觉效果上比扁椭圆形的红细胞略大。细胞核多分叶，通常为二分叶，核常偏位，染成紫红色。细胞质内充满被染成红色的杆状或纺锤形的颗粒，大小不均等的颗粒密集分布于细胞核周围，见图版I-C。
2.2.2 嗜酸性粒细胞
嗜酸性粒细胞在白细胞中占(1.03±1.96)%，细胞直径为(11.61±0.78)
μm。细胞常呈圆形或椭圆形，细胞核多分2叶或呈肾形，染成蓝紫色。此类细胞最大的特征为细胞质内充满大量圆形、均匀、排列紧密的橘红色嗜酸性颗粒，
