许多研究者在做氧气相关细胞实验时,通常不会关注培养基中溶氧的问题,而有文献表明:
- 把T75细胞培养瓶从常氧环境转移到2%的低氧环境后,需要约24小时培养基中的氧浓度才达到2%。
- 如在常氧环境中进行换液操作,仅4min后,培养基中氧浓度就从0%升至8.5%,而使其恢复到2%低氧则需约16h。

因此,在进行氧气相关的细胞实验时,不仅应关注细胞培养箱的O2浓度控制是否精确,还要关注培养基内溶解氧的浓度,否则可能极大地影响实验数据的可靠性和重复性。
Biospherix厂家有两点建议:
一、在进行低氧实验时,换液前应把培养基放置在低氧箱中进行预平衡。
二、换液及传代等工作尽量使用带氧气控制的操作台中完成,保持培养和操作环境的一致性,以提高低氧处理实验的成功率和可重复性。

工作原理:
Fiber Optic Dissolved Oxygen Sensor
This is accomplished by utilizing a special fiber optic oxygen
probe that has an inert fluorescent thin-film coating on the probe
tip.
A remote excitation light source will induce
fluorescence in this thin-film and the intensity of the
fluorescence is measured. The fluorescence quenching is then
converted as a function of absolute oxygen levels. Resolution is up
to 1 mmHg.
The probe tip is mounted on a stage above the culture vessel such
that it can be manually inserted into the media in any standard
culture dish or flask with either a small hole drilled in the cover
or topside surface (or dish without a cover), and retracted
manually as necessary. The probe tip must be removed from the
culture chamber manually when the chamber is removed from host
incubator to be transported to a glove box. The entire dissolved
oxygen sensing system will remain in the host incubator while the
culture chamber is removed, and the probe tip must be manually
inserted back into the stage when the culture chamber is returned
to the host incubator.
The stage can be mounted in a
micromanipulator at additional cost.
(In order to allow precise measurement and control of
oxygen at low levels over time in small volume cultures, the
dissolved oxygen sensors (fiber optic) used are designed to consume
no oxygen.)
